What exactly is a "Vertretungsprofessur" and how likely is it to get the position if you have one of these?Usually not worth it. Do not do it. They usually just want cheap teaching.
you do it if
1) you have never done serious teaching before, but you have already a good portfolio of publications
2) you don't do it if you still have not published enough, because it will eat up your time

I only did it, when I expected to get a call for a tenured professorship "cum spe".

I've heard enough stories to the contrary - but also stories where it did work, i.e., where the person who did all the work for the two years while the position was vacant did get the job in the end... the money is usually somewhat attractive (i.e. higher than the usual postdoc, iirc you get the non-civil servant salary of the position you are replacing). The main reason people do these jobs is because you can take an unpaid leave from your postdoc contract and thus prolong it.
I was on a postdoc contract and published well so I did it (Vertretungsprofessur) to signal that I can do the teaching/admin that comes along with a full prof position. Some time later I then got a full prof position (at a different place) and I do think the "Vertretung" was important to get there, though in substance it just ate away my time. I also saw this as a nice test for myself, i.e. if the full prof position in Germany can be squared with my doing research: I still managed to remain productive during the time of the "Vertretung", so in my case the answer was yes. I heared to often statements like "during the first two years as full prof you basically do not have time for research" and I just think for for people for whom this is true, they just never come back to do research afterwards.
Cologne is having talks for all three cluster positions.
